Ascletis Oral GLP-1 Hits Phase II Diabetes Endpoint as Phase III Obesity Program Enrolls

Topics: ASC30, Ascletis, oral GLP-1, type 2 diabetes trial, small molecule GLP-1, Phase II results, GLP-1 obesity pipeline

Ascletis Pharma has reported positive topline results from a U.S. Phase II study of ASC30, an oral small-molecule GLP-1 receptor agonist, in adults with type 2 diabetes — its second indication after obesity, reported while two global Phase III obesity trials are already enrolling.

What Happened

According to the company's Oct. 28 topline announcement, the 13-week randomized, double-blind, placebo-controlled study enrolled 100 adults with type 2 diabetes across multiple U.S. sites. Participants had a mean baseline HbA1c of 8.3%.

ASC30 met its primary endpoint: placebo-adjusted HbA1c reductions of roughly 1.6 to 1.7 percentage points at 12 weeks across three maintenance dose levels, statistically significant at p < 0.0001. On the secondary endpoint, placebo-adjusted body weight fell 3.8% to 4.4% at 13 weeks from a mean baseline of 93.7 kg, with no plateau observed.

Tolerability was the more distinctive claim. Ascletis reported zero discontinuations due to gastrointestinal adverse events during a weekly titration schedule, one discontinuation for any adverse event (reported as hiccup), no drug-related serious adverse events, and no hepatic signal — no elevations in ALT, AST, or total bilirubin during treatment.

Two caveats sit inside the data. The weight-loss effect is smaller than the 7.7% placebo-adjusted reduction Ascletis previously reported at Week 13 in its Phase II obesity study, which the company attributes partly to the diabetes population and a fast weekly titration it says will slow in Phase III. And weight was a secondary endpoint in a 100-person, 13-week study — a signal, not a durability finding.

Who Is Affected

No patients yet. ASC30 is investigational and unapproved in every market.

The readout matters to adults with type 2 diabetes who may eventually have a non-injectable option, to researchers tracking whether small molecules can approach the glycemic efficacy of peptide-based GLP-1s, and to the research-supply market — because ASC30 is not a peptide. It is a synthetic small molecule, a different regulatory and sourcing category from the compounded and research-grade peptides that dominate gray-market discussion.

Timeline

  • August 2026: Ascletis begins global Phase III obesity trials AURORA-1 and AURORA-2, targeting roughly 4,600 participants across the U.S., Europe, and Canada.
  • Oct. 28, 2026: Topline Phase II type 2 diabetes results announced.
  • Q3 2028: Phase III obesity topline data expected.
  • End of 2028: U.S. NDA submission planned for obesity; EMA application targeted for early 2029.

What This Means for Researchers

  • Placebo-adjusted is not absolute. The 1.6–1.7 point figure is the difference versus placebo, not the total fall in HbA1c.
  • Twelve weeks is short. It cannot speak to weight-loss durability, lean-mass retention, or long-term gastrointestinal tolerability.
  • Titration design drives the tolerability claim. The zero-GI-discontinuation result came from weekly titration; slower titration is expected to improve further, which is a projection.
  • Analytical methods do not transfer. Peptide identity and purity assays do not map cleanly onto a synthetic small molecule.

How to Verify Your Peptides

For anyone working with research peptides, the discipline stays the same whatever a vendor claims about the therapeutic class. Review our testing methodology, see what separates HPLC from LC-MS analysis, and check what identity, purity, and fill-weight testing actually confirms.

Safe Alternatives

Nothing here changes what is legally available today. FDA-approved GLP-1 products, where prescribed, remain the most tightly regulated option, and no generic versions of semaglutide or tirzepatide are approved. Our peptide research library and vendor directory profile suppliers with documented testing practices; background is available for semaglutide and tirzepatide.
